Virus Retention for Cell Culture Media

Virus-retentive filtration is a highly efficient method for reducing the viral risk of cell culture media. This filter targets both non-enveloped viruses such as MVM and vesivirus as well as large enveloped viruses such as MuLV. Virosart® Media is specifically designed for virus filtration of chemically defined cell culture media. Challenges in Virus Risk Reduction for Media Preparation: Multiple bioreactor contaminations reported over the past few years were caused by small, non-enveloped viruses such as MVM and vesivirus derived from raw materials. Our solution: Virosart® Media.

Virosart® Media is a virus-retentive filter specifically developed for upstream applications. The newly developed media filter overcomes the bottlenecks of today’s virus filters originally designed for downstream applications and provides a cost-effective upstream solution with the highest filter capacity for cell culture media.

Target molecule: The primary application of Virosart® Media is virus-retentive filtration of chemically defined cell culture media.

Virus clearance: Virosart® Media targets the removal of small, non-enveloped adventitious viruses (20 nm), such as MVM or vesivirus, from chemically defined cell culture media.

Positioning: Virosart® Media is used after the media preparation step before transferring the media to the bioreactor. This can be operated as inline filtration (e.g. perfusion) or batch filtration. The performance of Virosart® Media is independent of whether powdered or liquid media is used but can be significantly influenced by the media itself. Virosart® Media is optimized for chemically defined media. Protein transfer for mAbs and recombinant proteins is not specified for this filter.

Product Benefits

Robust virus clearance:
Virosart® Media provides effective removal of small non-enveloped viruses.

Short processing times: Based on the optimized Polyethersulfone membrane, Virosart® Media provides the highest flow rates and excellent capacity for cell culture media (e.g. ≥ 1000 L/m2 in 4 hours).

Smart process transfer: The hollow fiber format ensures the highest consistency and scalability from laboratory to process scale.

Cost efficiency: High packing density of the elements combines extremely low hold-up volume with minimal footprint requirements.

Ready to use: Sterile delivery ensures ease of use and quick installation of filter elements.

Ease of use: Virosart® Media is integrity-tested using a water-based diffusion test, e.g. based on Sartorius Stedim Biotech's Sartocheck® technology.

Regulatory compliance:

  • Each module is tested for integrity and water flow rate during manufacturing
  • Validated for removal of ≥ 4 LRV of small non-enveloped viruses using bacteriophage PP7
  • Designed, developed, and manufactured in accordance with an ISO 9001 certified Quality Management System
  • Meet or exceed the WFI quality standard requirements specified by the current USP
  • Non-pyrogenic according to USP Bacterial Endotoxins
  • USP Plastic Class Test VI
