Octet R8
The Octet® R8 label-free protein analysis system enables rapid, high-throughput, and accurate characterization of biomolecules. It is designed to support extended experimental run times during biotherapeutic development and integrates seamlessly into GxP-compliant laboratory environments.
The 8-channel Octet® R8 system performs quantification and kinetic analysis of up to 96 samples within 30 minutes to 2.5 hours, depending on the specific assay. An optional microplate evaporation cover minimizes sample volume loss and enables sample recovery after analysis.
The Octet® R8 system can be used for a wide range of applications, including kinetic analysis, titer determination of IgG and other proteins, reagent qualification, immunoassay development, bioprocess development, quality control analysis, crude antibody screening, epitope binning/mapping, ligand-binding assays, small molecule analysis, elucidation of cell signaling mechanisms, and infectious disease monitoring. Analyses can be performed using a single channel or up to eight channels, providing flexibility in sample throughput.

Applications
Kinetic and Affinity Characterization
The superior sensitivity of the Octet® R8 system enables measurement of both small and large molecules across a wide range of kinetic constants. By bringing the detection surface directly into contact with the sample, the system accurately measures kinetic constants while eliminating the need for microfluidics. The unique Dip and Read approach, combined with ready-to-use biosensors for most proteins, simplifies workflows and accelerates assay development. It also allows direct measurement of crude samples with minimal instrument maintenance.
Protein Quantification in Bioprocessing
Accurate and reproducible concentrations can be determined in as little as 2 minutes per sample or 32 minutes for a 96-well plate using a simple one-step assay. High sensitivity in quantification can be easily achieved by increasing read time and shaking speed. Even greater sensitivity, reaching sub-ng/mL levels, can be obtained with 2-step or 3-step assay formats, enabling faster and more sensitive automated measurement of contaminants such as host cell proteins and residual Protein A compared to ELISA. Process economics can be further improved through biosensor regeneration and reuse.
Quality Control (QC) and Manufacturing
The Octet® R8 system has been developed to operate reliably in regulated quality control (QC) environments. Octet® is widely used for method development and validation in lot release and in-process testing for ligand binding, concentration determination, and impurity analysis in both upstream and downstream processes.
