Octet R2
The Octet® R2 System characterizes protein-protein and protein-small molecule binding interactions for early-stage research and discovery. This versatile system also provides quantitative information by measuring active protein concentrations, even in complex mixtures such as cell culture supernatants and lysates. With the sensitivity required for small molecule analysis, the Octet® R2 instrument enables accurate determination of kinetic rates for interactions between small molecules as small as 150 Da and larger binding partners.
Budget-Friendly – Designed for academic and startup laboratories. When selecting a label-free assay system, low-volume users no longer need to choose between cost and performance.
Flexible second channel for reference measurements or additional samples – The unique 2-channel design features dedicated spectrometers for simultaneous reference measurements or increased capacity.
Grows with your research – Upgradeable to a 4-channel or 8-channel Octet® R Series system for higher throughput as your needs expand.

Benefits of the Octet® R2 Protein Analysis System
Kinetic Analysis
The Octet® R2 System monitors binding events in real time to determine association (Ka), dissociation (Kd), and affinity constants (KD). The system’s two channels can be used independently for screening purposes or together, pairing sample measurements with a dedicated reference for high-quality kinetic characterization.
Samples are loaded into standard microplates, eliminating fluidic line maintenance and increasing the number of samples that can be processed in a single assay. Predefined templates in Octet® BLI Discovery Software simplify assay setup and minimize training requirements. The software also provides users with a range of parameters and measurements for analyzing acquired data through a powerful yet intuitive interface.
Quantitative Measurements
The 2-channel Octet® R2 system directly detects the presence of specific proteins in solution with minimal interference from complex matrices. Accurate and reproducible concentrations can be determined in as little as two minutes per sample using a simple, single-step assay.
Sample Plate Cooling for Temperature-Sensitive Proteins
Sample plate temperature can be controlled between 15°C and 40°C, providing a reliable environment for kinetic studies involving temperature-sensitive proteins. Additional benefits include the ability to rapidly determine binding rate constants at multiple temperatures for estimating thermodynamic parameters.
Biosensor Re-Racking
The Octet® R2 system allows biosensors to be re-racked at the end of an experiment. This feature provides greater flexibility when loading ligands onto biosensors and reduces operating costs when biosensors are loaded with valuable target reagents.
Ease of Use
Robust technology combined with a simple, integrated user interface designed for biologists. This reduces the time required to learn and operate the instrument.
Applications
Large Molecule Characterization
With predefined templates available in Octet® BLI Discovery Software, the Octet® R2 system simplifies assay setup and minimizes training requirements. Octet® Analysis Studio then provides users with a comprehensive set of parameters and measurements to analyze experimental data through a powerful yet intuitive software interface.
Small Molecule Characterization
With the sensitivity required for screening small-molecule libraries, the Octet® R2 instrument enables measurement of interactions involving molecules as small as 150 Da for accurate determination of kinetic rates.
